Molecular beacons

Molecular beacons are hairpin-shaped oligonucleotides with the internally quenched fluorophore. Molecular beacons are designed in the way to remain an intact stem-loop structure during primer elongation, while after each cycle they bind to amplicon to quantify the target. Also, you can order molecular beacons with additional chemical modifications to increase thermal stability and specificity.

  • Molecular beacons form a stem-loop structure that bring a 5′ fluorescent dye and a 3′ quencher together. Stem can be universal for all probes, while the loop is able to hybridize to the target DNA with high specificity. As a result, the reporter dye and quencher are separated sufficiently to induce the increase of fluorescence. During amplification these probes only bind to DNA without degradation by Taq polymerase.
